技术桥梁,靶向链接——PROTAC Linker助力新药研发-百灵威

当前临床治疗中普遍采用的药物多是基于小分子设计的,它们通过“占据驱动”的机制来抑制蛋白质的功能,从而达到治疗疾病的目的。

然而,与传统的小分子抑制剂和拮抗剂不同,PROTAC(蛋白降解靶向联合体)技术(图1)通过诱导致病靶蛋白的降解,开辟了药物研发的新路径。近年来,这一技术发展迅猛,已成为新药开发领域的重要工具。

PROTAC Linker(PROTAC 连接子)的最佳长度12个碳到20 个碳不等,在 PROTAC 构建中起着重要作用,用来连接 PROTAC 的两个功能基团,靶蛋白配体和E3 连接酶配体。较常用的接头是PEG、烷基链和烷基醚。
