肖海华课题组科研成果-抗肿瘤四价铂类纳米药物-百灵威

    肿瘤免疫抑制微环境严重削弱机体抗癌能力,降低药物疗效,加速肿瘤恶化转移。其中,骨髓来源的抑制性细胞(MDSCs)通过抑制细胞毒性T淋巴细胞(CD8+ T细胞)激活,阻碍免疫系统对肿瘤的识别与清除,成为肿瘤进展的关键因素。因此,开发能同步抑制MDSCs浸润、激活CD8+ T细胞的疗法极具临床意义。

    中国科学院化学研究所肖海华课题组深耕生物医用材料领域,在四价铂类纳米药物抗癌研究中取得重要成果。团队设计合成双亲性高分子PolyRA与四价铂前药Oxa-c16,组装成 PRA@Oxa-c16 纳米粒。该纳米粒可有效抑制MDSCs向肿瘤部位聚集,诱导免疫原性细胞死亡,显著增强CD8+ T细胞的抗癌活性。联合α PD-L1抗体使用时,不仅能清除小鼠皮下肿瘤、抑制肺转移,还大幅降低肿瘤复发风险,为肿瘤免疫治疗提供了新策略。
